Soybean (Glycine max (L.) Merr.) oil is an important source of vegetable for supporting the human diet. However, high level polyunsaturated fatty acids in natural soybean renders unstable and thus susceptible to development unpalatable flavors trans acids. Therefore, reducing content increasing monounsaturated a longstanding target breeding. varieties with oleic acid are rare germplasm resources, which introduces substantial difficulties cultivation high-oleic-acid soybeans. In this study, CRISPR/Cas9-mediated gene-editing technology was used create targeted knockout desaturase encoding genes GmFAD2-1A GmFAD2-1B that contribute formation We obtained fad2-1a, fad2-1b, fad2-1a/fad2-1b homozygous mutants using two sgRNAs. found increased from 11% 40-50% fad2-1a fad2-1b 85% mutants. also generated transgene-free double conferred higher acid, mutant had no adverse phenotyping compared wild type. Our study provided new materials selection breeding varieties.
